Product Description

Norgen’s Urine DNA Concentration, Preservation and Isolation Kit is an all-in-one solution designed for:

  1. Rapid and simple urine concentration;
  2. Preservation of DNA from concentrated urine samples at ambient temperature;
  3. Isolation of high quality, inhibitor-free urineDNA within a laboratory setting.

Norgen'sUrine DNA Concentration, Preservation and IsolationKit contains Individual Urine Concentration and Preservation Devices, as well as the required columns and reagents for the subsequent laboratory isolation of the urine DNA from the preserved samples. Each of the Individual Urine Concentration and PreservationDevices consists of2 keycomponents: (1) a Urine Concentration Tube, and(2) Norgen’s Urine Preservative contained within sealed squeezable ampules.

Briefly, urine samples are collected directly by donors in any urine collection container, and then the urine istransfered into the Urine Concentration Tube up to the indicated fill line. The urine is then mixed by shaking such that theliquid and resininside the Urine Concentration Tube are in complete resuspension with the urine sample. All the macromolecules present within the urine will bind to the resin in the presence of the specially formulated buffer located within the Urine Concentration Tube. The resuspended resin is then allowed to settle by gravity to the bottom of the Urine Concentration Tube. When the entire contents of the Urine Concentration Tubeare completely precipitated, the urine supernatant is then decanted and removed. The Urine Preservative is then added to the precipitated resin and again the tube is shaken to allow for the Urine Preservative and the resin to become fully mixed and resuspended. If desired the Urine Concentration Tube can now be labelled using the providedID Label, or alternatively the unique identifier code on the tube can be used for tracking of the sample. Thepreserved urine sample can now be sent to the lab for DNAisolation and analysis. Shippingaccessories are available separatelyto allow for safe and easy shipping of the preserved urine sample.

Urine DNA that has been concentrated and preserved usingthe Urine Concentrationand PreservationDeviceis stable for years at room temperature. This kit is ideal for concentrating, preserving, shipping and isolating DNA samples for epidemiological and population studies.

URINE PRESERVATIVE
Norgen’s Urine Preservative is an aqueous storage buffer that prevents the growth of Gram-negative and Gram-positive bacteria and fungi, and also inactivates viruses allowing the resulting non-infectious urine samples to be handled and shipped safely. In addition, the buffer eliminates the need to immediately process or freeze samples and allows the samples to be shipped to centralized testing facilities at ambient temperature. The components of the buffer allow samples to be stored for over 24 months without any detectable DNA degradation.

DNAISOLATIONFROMCONCENTRATED, PRESERVEDURINESAMPLES
Purification is based on spin column chromatography using Norgen's proprietary resin as the separation matrix. Briefly, concentrated and preserved urine samples are lysed using Proteinase K and Pronase. The lysed urine samples are then mixed with Binding Buffer and the resulting slurry with bound urine DNA is loaded onto a provided Mini Filter Spin Column. Next, the bound urine DNAis washedto removeany remaining proteins or other impurities. Lastly, the purified total urine DNA is eluted into 50-300 µL of the provided Elution Buffer.

The kit allows for the removal of highly concentrated salts, metabolic wastes and proteins to allow for the isolation of high quality, concentrated DNA. Preparation time for a single sample is approximately 30 minutes. The purified urine DNA is compatible with most of the molecular biological applications such as PCR, q-PCR and DNA fingerprinting

Figure 2. Stability of DNA in Norgen's Urine Preservative. The main component of Norgen's Urine DNA Concentration, Preservation and Isolation Kit is the Urine Preservative. Urine samples were collected from numerous donors andpooled, and then the urine was aliquoted into 30 mLsamples and concentrated as per the provided protocol.Norgen’s Urine Preservative was added to each of the concentrated urine samples. The preserved DNA was stored at -20°C, 4°C, room temperature and 55°C for up to 6 months. Urine DNA was subsequently isolated at time 0, 1 month, 2 months, 4 months and 6 months from the concentrated and preserved urine sample. The DNA isolated from the concentrated and preserved urine samples stored at -20°C, 4°C, room temperature and 55°C at the various time points was used as the template in a real-time PCR reaction to detect the 5S gene. As it can be seen, all the DNA samples were successfully amplified. Furthermore, the quality of the concentrated and preserved urine DNA stored at all 4 temperatures did not change from 0 months to 6 months, as indicated by the fact that the Ct value remained consistent. Theurine samples stored at 55°C can allow us to extrapolate results based on accelerated-aging, and therefore the samples stored at 55°C for 6 months show equal stability to concentrated urine preserved for 2.5 years at room temperature (24°C). Therefore, based on the successful PCR results for samples stored at 55°C shown above, concentrated urine samples stored in Norgen’s Urine Preservative are stable at room temperature for up to 2.5 years.

Twenty different urine samples were collected, concentrated and preserved using Norgen’s Urine DNA Concentration, Preservation and Isolation Kit, and were stored at room temperature for 1 day. DNA was subsequently purified according to the provided protocol and eluted into 150 µL, and the DNAconcentration was determined using a NanoVue PlusTM (GE Healthcare). The total averageurine DNA concentration fromthe 20 different urine samples processed using Norgen’s kit was 141 ng/µL. Figure3 shows the DNA concentration from the 20 different 30 mL concentrated and preserved urine samples.

Twenty different urine samples were collected, concentrated and preservedusing Norgen’s Urine DNA Concentration, Preservation and Isolation Kit, and were stored at room temperature for 1 day. DNA was subsequently purified according to the supplied protocol, and the DNA purity (OD260/280) was determined using a NanoVue PlusTM (GE Healthcare). The average purity of the isolatedurine DNA was 1.56+0.16.

Urine samples were collected from 20 donors using Norgen’s Urine DNA Concentration, Preservation and Isolation Kit, and urine DNA was subsequently isolated from the 20 samples using the supplied protocol. Two microliter from eachurine DNA sample was then used as the template in a real-time PCR reaction using 2X SYBR Green/Fluorescein qPCR Master Mix (Norgen Biotek, Cat# 28109) and 5S specific primers. All 20 samples showed successful amplification, indicating the high quality of the purified DNA.

Ten urine samples were collected using Norgen's Urine DNA Concentration, Preservation and Isolation Kit, and the urine DNA was isolated according to the supplied protocol. Two microliter of each of the isolated DNA samples was used as the template in an end-point PCR reaction to amplify the GAPDH gene. All of the ten urine DNA samples were successfully amplified indicating that no PCR inhibitors were co-purified with the DNA (Lanes 1 to 10). Urine DNA samples purified using Norgen's kit are therefore compatible with sensitive downstream applications. Lane M is Norgen's FastRunner DNA Ladder.

SHELF LIFE AND HANDLING
This kit contains ready-to-use Pronase and Proteinase K which is dissolved in a specially prepared storage buffer. The Proteinase K should be stored at 4°C upon arrival (stable for 2 years). All other kit components should be kept tightly sealed and stored at room temperature (15 – 25 °C) for up to 2 years without any reduction in kit performance.
