Product Description

Norgen’s Bacterial Genomic DNA Isolation Kit is designed for the rapid preparation of genomic DNA from a few cells up to 2 x 109 viable bacterial cells (between 0.5 and 1 mL of culture).

Purification is based on spin column chromatography. Briefly, the bacteria are pelleted and lysed,and the released bacterial genomic DNA is bound to Norgen's column in the presence of a Binding Solution (BIND). Under these conditions only the genomic DNAwill bind to Norgen’s resinwhile most of thecontaminating RNA and cellular proteinaceous components are removed in the flowthorugh. The bound DNAis then washedto remove any remaining impurities(WASH). Lastly, the purifiedbacterial genomic DNAiseluted into100-200 µL of the provided ElutionBuffer or water (ELUTE). The purified DNA is suitable for many downstream applications, includingPCR, qPCR and Southern blotting.The Bacterial Genomic DNA Isolation Kit allows for the isolation of genomic DNA from both Gram negative and Gram positive cultures, includingE. coliandB. cereus. Typical purification yields are up to 20 µg of genomic DNA, but will vary depending on the cell density of the bacterial culture and the bacterial species.

The high yield of the Norgen’s Bacterial Genomic DNA Isolation Kit is illustrated by purifying genomic DNA from 1 mL overnight culture (1 x 109 cells) of both a Gram positive (B. cereus) and a Gram negative strain (E coli), and comparing the yield with two major competitors.The quantification of the DNA yield was performed by resolving 5 µL of the 200 µL of eluted DNA on a 1X TAE, 0.9% agarose gel followed by densitometry. With both types of bacteria, Norgen’s kit was found to give a higher recovery than the competitor’s kits.

Storage Conditions and Product Stability
All solutions should be kept tightly sealed and stored at room temperature. These reagents should remain stable for at least 1 year in their unopened containers. The Proteinase K should be stored in aliquots at -20°C upon reconstitution. This product is stable at room temperature in its lyophilized form.
